Cell cycle analysis is a widely employed biological parameter for the assessment of the toxicity of chemical compounds in both the drug discovery industry and biomedical research. Interruption of cell proliferation and disturbance of the cell cycle represent highly sensitive markers of cytotoxicity. Analysis of the effect of lead compounds on DNA synthesis, including mitotic arrest or apoptosis are commonly studied for the development of novel compounds for a number of therapeutic areas such as oncology, cardiovascular, and metabolic disorders.
Cell cycle analysis requires accurate determination of DNA content, and is routinely performed using flow cytometry, which is slow. By contrast, TTP Labtech’s acumen Cellista allows determination of cell number, cytotoxicity and cell cycle analysis in a single, fast, dual laser read.
Its advantages include:
- uniform illumination over the whole well (flat field illumination) which gives better data on assays such as cell cycle, where there is only a 2-fold difference from background
- a simple dye-based method can be enhanced by multiplexing with other markers of interest (e.g. mitotic index). This method includes Hoeschst and propidium iodide dyes which offer a very cost-effective cell labelling in comparison to antibody-based approaches.
- in situ analysis means a workflow that is easier to automate than flow cytometry and offers improved data quality that maintains cell morphology
